(A) Cells are treated with the selected compounds (AOx) and the dichlorofluorescein diacetate molecule, DCF-DA, which is absorbed by cells by passive diffusion and deacetylated by cell esterases to non-fluorescent DCFH. (B) DCFH is rapidly oxidized by free radicals, produced by 2,2'-Azobis [2-methylpropionamide] dihydrochloride, AAPH, a peroxyl radical initiator. Free radicals lead to the oxidation of DCFH to DCF which emits fluorescence proportional to the level of oxidation. Under these conditions, a decrease in the fluorescence signal compared to untreated cells indicates the antioxidant capacity of the tested compound, as antioxidants react with peroxyl radicals preventing the formation of fluorescent DCF dichlorofluorescein.
